Published research on Hydroxychloroquine sulfate

5 peer-reviewed publications on Hydroxychloroquine sulfate in oncology research, clinical work first, then laboratory studies. Each entry reports what the authors stated; follow the link for the full paper.

This page is a reading list of peer-reviewed publications, compiled for information. It is not a statement of indications, a recommendation or medical advice. Results from cell and animal studies do not translate into clinical use. Medsuvac products are supplied for their registered purposes; clinical decisions belong to a qualified physician.
  1. Clinical trial

A Randomized Phase II Preoperative Study of Autophagy Inhibition with High-Dose Hydroxychloroquine and Gemcitabine/Nab-Paclitaxel in Pancreatic Cancer Patients

Zeh HJ et al., Clinical Cancer Research, 2020

In 64 patients with resectable pancreatic adenocarcinoma, adding hydroxychloroquine to preoperative gemcitabine and nab-paclitaxel improved histopathologic response and showed evidence of autophagy inhibition and immune infiltration, while overall survival and relapse-free survival did not differ between arms.

  2. Clinical trial

Effect of Gemcitabine and nab-Paclitaxel With or Without Hydroxychloroquine on Patients With Advanced Pancreatic Cancer: A Phase 2 Randomized Clinical Trial

Karasic TB et al., JAMA Oncology, 2019

In 112 patients with metastatic pancreatic cancer, adding hydroxychloroquine to gemcitabine and nab-paclitaxel did not improve overall survival at 12 months (41% vs 49%), although the overall response rate was higher in the hydroxychloroquine group (38.2% vs 21.1%).

  3. Clinical trial

Phase I trial of hydroxychloroquine with dose-intense temozolomide in patients with advanced solid tumors and melanoma

Rangwala R et al., Autophagy, 2014

In 40 patients with advanced solid tumors, the combination of hydroxychloroquine and dose-intense temozolomide was well tolerated, no maximum tolerated dose was reached, autophagic vacuole accumulation was observed in patient cells, and partial responses or stable disease occurred in a subset of melanoma patients.

  4. Clinical trial

Combined autophagy and proteasome inhibition: a phase 1 trial of hydroxychloroquine and bortezomib in patients with relapsed/refractory myeloma

Vogl DT et al., Autophagy, 2014

In 25 patients with relapsed or refractory myeloma, combining hydroxychloroquine with bortezomib produced no protocol-defined dose-limiting toxicities, and of 22 evaluable patients 3 had very good partial responses, 3 had minor responses and 10 had stable disease.

  5. Review

Outcome of early clinical trials of the combination of hydroxychloroquine with chemotherapy in cancer

Poklepovic A et al., Autophagy, 2014

The authors summarized five phase I trials combining hydroxychloroquine with vorinostat, temsirolimus, temozolomide or bortezomib and stated that these studies provided insights into tolerability of the combinations and guidance for the design of future trials.
