  1. Clinical trial

Propranolol monotherapy in angiosarcoma: a window-of-opportunity study (PropAngio)

Embaby A et al., European Journal of Cancer, 2024

In 14 patients with angiosarcoma treated with propranolol monotherapy for 3 to 6 weeks before standard care, two patients showed clinical response and none showed histologic response, and the trial did not demonstrate clinical efficacy of propranolol monotherapy.

  2. Clinical trial

Phase I Clinical Trial of Combination Propranolol and Pembrolizumab in Locally Advanced and Metastatic Melanoma: Safety, Tolerability, and Preliminary Evidence of Antitumor Activity

Gandhi S et al., Clinical Cancer Research, 2021

In a 3 + 3 dose-escalation study of nine patients with treatment-naive metastatic melanoma, propranolol combined with pembrolizumab produced no dose-limiting toxicities and an objective response rate of 78%.

  3. Clinical trial

Preoperative beta-Blockade with Propranolol Reduces Biomarkers of Metastasis in Breast Cancer: A Phase II Randomized Trial

Hiller JG et al., Clinical Cancer Research, 2020

In a triple-blind placebo-controlled trial of 60 breast cancer patients, 7 days of preoperative propranolol downregulated mesenchymal gene expression in the primary tumor without affecting epithelial gene expression and was associated with changes in intratumoral immune cell infiltration.

  4. Observational

Propranolol for Off-label Treatment of Patients With Melanoma: Results From a Cohort Study

De Giorgi V et al., JAMA Oncology, 2018

In a prospective cohort of 53 patients with stage IB to IIIA melanoma, off-label propranolol use at diagnosis was inversely associated with recurrence after adjustment for prognostic factors (hazard ratio 0.18, 95% CI 0.04 to 0.89).
